First make sure the call was from your bank.
Call them back using the phone number from your card or statement.
They should be able to connect you to the Fraud Department.
If the call is legitimate, then there's probably something about the charge that is suspicious.
That may be the nature of the charge, where it was from, who it was from, or difference compared to your usual charging pattern.
Determine if the charge is legitimate, one you actually agreed to.  You may also dispute charges you find on your statements, but be sure to file disputes with your bank within 60 days of the statement date of the statement showing the dispute, to fit within the bank dispute window under FRB Reg. E or FCBA.

Be sure to dispute promptly, but always follow up IN WRITING.  Since bank disputes must be filed within 60 days of the statement date, sending a dispute by certified mail ensures that you have a record proving a timely dispute.
If the charge is fraudulent, insist on receiving a fraud affidavit, sign and send it in.
Keep a copy of all correspondence, and keep notes logging all calls.
If your card or account number has been used for fraudulent charges, you should block the card number or shut down the account to prevent further charges.  If you don't, you might get stuck for further fraudulent charges, particularly if you miss disputing them.
